Floating leaf disk assay measuring the effect of temperature on photosynthetic rate in Spinacia oleracea.

Designed and conducted an experiment investigating how temperature (20–60 °C) affects the photosynthetic rate of spinach leaf disks in a sodium bicarbonate solution. Photosynthetic rate was measured indirectly via disk resurfacing time and the ET50 metric. Statistical analysis using one-way ANOVA (F = 4.43, p = 0.0023) and Tukey HSD post-hoc tests identified 30 °C as the optimum temperature, with enzymatic denaturation of RuBisCO activase driving decline at higher temperatures.
